Ox-low density lipoprotein (ox-LDL) increased the susceptibility of primary nephrotic syndrome (PNS) by regulating the Treg/Th17 ratio via the MEG3 signaling

Introduction Patients with primary nephrotic syndrome (PNS) were reported to exhibit the evident imbalance between number of Th17 and Treg cells in their peripheral blood monocytes (PBMCs), which might be immunological basis disease. Material methods 40 PNS patients 42 healthy individuals recruited this study. FCM assay was used observe levels cells. Results The Treg/Th17 ratio evidently decreased patients. MEG3 RORγt increased group, while miR-17, miR-125a FOXP3 mRNA reduced group. Moreover, IL-6 IL-1β highly Ox-low density lipoprotein (ox-LDL) treatment significantly mRNA/protein decreasing miR-125a, THP-1 cells, transfection siRNA partly alleviated dysregulation expression MEG3, relevant miRNAs mRNAs induced by ox-LDL. Also, miR-17 upon successful but promoted inhibited. Conclusions Our results demonstrated that ox-LDL could promote inflammatory response via activating signaling. activation MEG3/miR-125a/FOXP3 axis MEG3/miR-17/RORγt respectively lead imbalance, resulted up-regulated levels, thus increasing susceptibility PNS.
